Zidovir® manufacturer is Cipla. Its active chemical compound is Zidovudine that belongs to the drug class called synthetic guanosine nucleoside reverse transcriptase inhibitors (NRTIs).


- treatment of human immunodeficiency virus (HIV-1) infection in combination with other anti-retroviral drugs;

- prevention of the transmission of HIV during childbirth.

MOA (mechanism of action):

Zidovudine is a structural analog of thymidine, a prodrug that must be phosphorylated to its active 5-triphosphate metabolite, zidovudine triphosphate (ZDV-TP). It inhibits the activity of HIV-1 reverse transcriptase (RT) by terminating DNA chain after incorporation of the nucleotide analogue. It competes with the natural substrate dGTP and incorporates itself into viral DNA. It is also a weak inhibitor of cellular DNA polymerase. Zidovudine isn't active against HIV-2.


The usual dose for patients weighing 25 kg or more is one tablet of 300 mg twice a day. Each dose should be taken approximately 12 hours apart.

Swallow tablets whole with water or another drink.

Zidovir® can be taken with or without food.

Use other formulations with Zidovudine for dosing in children weighing less than 25 kg.


HIV can be transmitted through sexual contact with HIV positive partner, blood (e.g., sharing needles, handling blood instruments, receiving blood transfusions, blood products, or organ/tissue transplants that were infected with HIV), childbirth, toiletries (razors and toothbrushes). Therefore, both HIV-positive and HIV-negative individuals are advised to have any kind of sex using a latex or polyurethane condom, avoid sharing needles or other things like toothbrushes or razors. It is important to discuss with doctor the precautions needed to avoid infecting other people as well as take precautions not to get infected.

Take special care if you have moderate or severe liver disease, you have ever had liver disease, including hepatitis B or C, you are seriously overweight (especially if you are a woman), you have severe kidney disease.

Consult doctor before taking if you are taking phenytoin (for treating epilepsy), methadone (as a heroin substitute). Zidovudine speeds up the removal of methadone from the body. If you are taking methadone, you will be checked for any withdrawal symptoms. Your methadone dose may need to be re-elevated.

Women who are HIV-positive must not breastfeed because HIV infection can be passed on to the baby in breast milk.

Some people who take Zidovir® may experience:

- Anaemia (low red blood cell count) and neutropenia/leucopenia (low white blood cell count).

- Lactic acidosis (a build up of lactic acid in the body), together with an enlarged liver. Symptoms are deep, rapid breathing, drowsiness, and non specific symptoms such as nausea, vomiting and stomach pain.

- Immune Reconstitution Syndrome (immune response to fight infections that may have been hidden in the body).

- Loss of body fat may occur in patients receiving Zidovir®.

- Bone disease osteonecrosis (death of bone tissue).

Missing a dose

If you miss a dose you should take it as soon as you remember about your missing. If it is the time for the next dose you should continue your regular dosing schedule. Do not take a double dose of Zidovir® to compensate the missed dose.


If an overdose occurs and you are not feeling well, stop taking the medication immediately. You should seek emergency medical attention or contact your healthcare provider immediately.


Store at room temperature between 15 and 30 degrees C (59 and 86 degrees F) and away from excess moisture and heat. Throw away any unused medicine after the expiration date. Keep out of the reach of children.

Side effects

Common and uncommon side effects associated with Zidovir® are:

  • headache
  • nausea
  • vomiting
  • muscle pain
  • anaemia
  • leucopenia
  • neutropenia
  • increase in liver enzymes and bilirubin in the blood
  • anxiety
  • depression
  • insomnia
  • nail and skin pigmentation
  • hives
  • sweating
  • fat accumulation in the liver
  • increased urinary frequency

Side effects occurrence does not only depend on the medication you are taking, but also on your overall health and other factors.


Zidovir® is contraindicated in patients who are allergic to Zidovudine or any of the other ingredients of Zidovir®.

Do not take Zidovir® if you have or have nad very low red blood cell count (severe anaemia) or very low white blood cell count (neutropenia), kidney disease or liver disease (such as hepatitis), peripheral neuropathy, pancreatitis.

Zidovir® is contraindicated if you are taking other medications like stavudine, ribavirin, valproic acid, fluconazol, probenecid, pentamidine, dapsone, pyrimethamine, cotrimoxazole, amphotericin, flucytosine, ganciclovir, valganciclovir, interferon, vincristine, vinblastine and doxorubicin.

Consult your doctor before taking Zidovir® if you are pregnant or plan to have a baby.

A: Zidovir® is a potent medication used to treat HIV-1 infection in conjunction with other anti-retroviral drugs. Zidovir® tablets may improve the condition by increasing the count of CD4 cells (white blood cells) that help fight infection. Zidovir® is not a cure for HIV-1 infection.
